Transfection of bovine spermatogonial stem cells in vitro
Authors
Abstract:
Spermatogonial stem cells (SSCs) are the only stem cells in adults that can transfer genetic information to the future generations. Considering the fact that a single SSC gives rise to a vast number of spermatozoa, genetic manipulation of these cells is a potential novel technology with feasible application to various animal species. The aim of this study was to evaluate enhanced green fluorescent protein (EGFP) gene transfection into bovine SSCs via liposome carrier and assess the best incubation day in uptake exogenous gene by SSCs. Transfection efficiency of EGFP gene with lipofectamine 2000 was determined in days following each three day of transfection (day 4, 6 and 8 of the culture) by fluorescent microscope. Results showed that the transfected cells through lipofection increased significantly (P

Biomanipulation of bovine spermatogonial stem cells
The field of spermatogonial stem cell (SSC) technologies provides tools for genetic improvement of cattle herds and multiple opportunities for research. Spermatogonial stems cells belong to the male germ line and as such have high developmental potential, which offers many possibilities for transfer of relevant genetic traits across herds in a timely manner. Type A spermatogonia include a very ...

background: niche cells, regulating spermatogonial stem cells (sscs) fate are believed to have a reciprocal communication with sscs. the present study was conducted to evaluate the effect of ssc elimination on the gene expression of glial cell line-derived neurotrophic factor (gdnf), fibroblast growth factor 2 (fgf2) and kit ligand (kitlg), which are the main growth factors regulating sscs deve...
full textIn Vitro Culture-Induced Pluripotency of Human Spermatogonial Stem Cells
Unipotent spermatogonial stem cells (SSCs) can be transformed into ESC-like cells that exhibit pluripotency in vitro. However, except for mouse models, their characterization and their origins have remained controversies in other models including humans. This controversy has arisen primarily from the lack of the direct induction of ESC-like cells from well-characterized SSCs. Thus, the aim of t...
